Measuring Bacterial and Fungal Substrate-induced Respiration in Dry Soils
نویسنده
چکیده
The substrate-induced respiration inhibition (SIRIN) method of Anderson and Domsch for partitioning bacteria1 and fungal contributions to soil respiration was modified for application to dry soils. This new method also provided a comparative basis when measuring SIRIN in soils of different moisture contents. Soil was incubated under optimum moisture conditions (55% water-filled pore space) to maximize microbial activity and to ensure homogeneous incorporation of substrate and inhibitors into soil. Soil samples were packed to a uniform bulk density prior to measurement of CO2 evolution by gas chromatography. Glucose (3 mg g ‘) was added together with streptomycin (0.5 or 1 .O mg g‘) and/or cycloheximide (15 mg gg ‘) for selective respiratory inhibition. The procedure included conditioning for 16 h at 4”C, followed by 1.5-h equilibration and 2-h incubation. The method yielded consistent and reproducible CO, respiration measurements for soils from a semi-arid region having gravimetric moisture contents ranging between 7.5 and 23.2%. Method sensitivity was not sufficient to detect variations in the fungal-to-bacterial ratio due to management practice for the soil under study. Measured fungal-to-bacterial ratios of 29: 1 and 15:1, for conventionally and no-till managed soil, were not significantly different at a probability level of 5%. Copyright

Sensitivity of soil respiration and microbial communities to altered snowfall
Winter respiration is a quantitatively important, yet variable flux of carbon dioxide (CO2) from soils to the atmosphere. Variability in winter soil respiration may be influenced by the effects of snowfall on microbial communities and their metabolic activities. In this study, we evaluated the importance of snowpack depth on soil respiration and microbial communities in a temperate deciduous fo...
متن کاملRelationships Between land Use and Arbuscular Mycorrhizal (AM) Spore Abundance in Calcareous Soils
This study was conducted to determine soil properties that correlate with the arbuscular mycorrhizal fungal (AMF) spore numbers in semiarid calcareous soils of Hamadan province in northwestern of Iran. Soil samples from six sites managed differently were collected from a 0 to 30 cm depth. The results showed that land use and management systems had a significant effect on AMF spore number in soi...

Extended abstract Introduction Topography is one of the effective factors in soil formation and development. Topographical features such as slope aspect and position, by affecting soil temperature, evaporation capacity, soil moisture content, soil organic matter, precipitation, movement, and accumulation of soil solution can impress soil microbial properties. For investigating the ...
متن کاملImpact of metal pollution and Thlaspi caerulescens growth on soil microbial communities.
Soil microorganisms drive critical functions in plant-soil systems. As such, various microbial properties have been proposed as indicators of soil functioning, making them potentially useful in evaluating the recovery of polluted soils via phytoremediation strategies. To evaluate microbial responses to metal phytoextraction using hyperaccumulators, a microcosm experiment was carried out to stud...
متن کاملA Simple Method for Measuring Co2 in a Continuous Air-flow System: Modifications to the Substrate-induced Respiration Technique
A simple, inexpensive method for measurement of respiration (CO, evolution) under a continuous air-flow was calibrated against an i.r. gas analyzer. Regression equations were developed to apply the method to general soil respiration measurement in the laboratory. The substrate-induced respiration technique was modified. By using 120% PWHC as a standard soil water condition during incubation, in...
